Yes, acrylic paint is waterproof. However, it is hard to imagine the paint being applied from beneath, through the floor. It is not the same as using waxes or oils. Any acrylic paint with lacquers in it is better in this regard (especially water-based). The reason is the paint has many layers, and the water that enters a hole must make an opening large enough for the paint to dissolve. Also, it goes into a larger hole than using wax or oil would do; if the paint is put in an opening smaller than the hole it will be difficult to remove the excess paint.

It is hard to predict the specific quality of the coating because the different lacquers all have their own properties. The manufacturers use a technique called “fractional dispersion” of the layers so that each layer can remain intact without compromising the rest. But many layers of paint will remain intact, and the paint will remain color-treated regardless of what kind of coating it is applied on top of, for instance, to a window.
There is one type of colored acrylic paint that is better than others: the one that contains all the colors but the majority of the pigment, known as DMC/Chinacolor. In most cases these types of paints are waterproof because it is possible to get the paint into the crack and get to what most likely is water through the cracks (or just in the paint’s recesses). There is a difference between two coats, which can occur even if one is waterproof (in which case the second may be too).

Generally, the thicker the layer of paint (the one that includes lacquer), the longer it will take to be covered with it. If it takes just three layers of paint, then you have a reasonably good one. However, if it takes six or eight layers, then you most likely need a second coat.
If it takes 10-12 layers, then you are usually in for a second one. Since the layers are not uniformly spaced, the second coat generally adds to the length of the paint job. If it takes 16 or 18 coats, then it is going to be very challenging
